Using a stock fuel rail do you need to remove the stock fuel pressure reg in order to fit the aftermarket adjustable item?

No just take the vacuum line off and plug into new reg, put the new one inline of the return fuel line.

Yes

Depending on how you go about mounting it, you might have to get Fuel Rail adapters, unless the FPR you bought bolts straight to the rail.

I thought they had to go on the return line so they can build pressure up for the injectors?

correct, they go between the rail and return line to the tank

No just take the vacuum line off and plug into new reg, put the new one inline of the return fuel line.

But then the fuel pressure is fixed at the atmospheric setting of the stock FPR. It won't matter how much boost or vacuum you have, the rail pressure will be constant.
