Experiment 2:

Analysis of Genome Structure

Abstract:

Organisms have a DNA genome (with the exception of some RNA Viruses). These genomes come in different sizes and shapes. Some organisms have linear chromosomes consisting of single or double stranded DNA whilst others have circular genomes. The genome of the bacterium Escherichia coli(E.coli) is 4.2 x 106 base pairs (bp) in size, the fruit fly Drosophila melanogaster is 1.4x108 bp and the human genome is 3.3x109bp. Difference in genome structure these fragments can be analysed by gel electrophoresis.

DNA digesting enzymes may be used to generate a series of DNA fragments whose numbers and length depends on the size and shape of the genome. These fragments can be analysed by gel electrophoresis.
